Emerin Represses STAT3 Signaling through Nuclear Membrane-Based Spatial Control

چکیده

Emerin is the inner nuclear membrane protein involved in maintaining mechanical integrity of membrane. Mutations EMD encoding emerin cause Emery–Dreifuss muscular dystrophy (EDMD). Evidence accumulating that regulation specific gene expression associated with this disease, but exact function has not been fully elucidated. Here, we show downregulates Signal transducer and activators transcription 3 (STAT3) signaling, activated exclusively by Janus kinase (JAK). Deletion mutation experiments lamin-binding domain essential for inhibition STAT3 signaling. interacts directly co-localizes knockdown induces target genes Bcl2 Survivin to increase cell survival signals suppress hydrogen peroxide-induced death HeLa cells. Specifically, downregulation BAF or lamin A/C increases suggesting correct-localized emerin, assembling A/C, acts as an intrinsic inhibitor against In C2C12 cells, gene, Pax7, abnormal myoblast proliferation muscle wasting skeletal homeostasis. Our results indicate signaling inducing retention delaying This mechanism provides clues etiology emerin-related may be a new therapeutic treatment.

A novel role for the nuclear membrane protein emerin in association of the centrosome to the outer nuclear membrane

The type II inner nuclear membrane protein emerin is a component of the LINC complex that connects the nuclear lamina to the actin cytoskeleton. In emerin-null or -deficient human dermal fibroblasts we find that the centrosome is detached from the nucleus. Moreover, following siRNA knockdown of emerin in wild-type fibroblasts, the centrosome also becomes detached from the nucleus. We show that ...

Transforming Growth Factor-β1 Signaling Represses Testicular Steroidogenesis through Cross-Talk with Orphan Nuclear Receptor Nur77

Transforming growth factor- β1 (TGF-β1) has been reported to inhibit luteinizing hormone (LH) mediated-steroidogenesis in testicular Leydig cells. However, the mechanism by which TGF-β1 controls the steroidogenesis in Leydig cells is not well understood. Here, we investigated the possibility that TGF-β1 represses steroidogenesis through cross-talk with the orphan nuclear receptor Nur77. Nur77, ...

The nuclear envelope LEM-domain protein emerin

Emerin, a conserved LEM-domain protein, is among the few nuclear membrane proteins for which extensive basic knowledge--biochemistry, partners, functions, localizations, posttranslational regulation, roles in development and links to human disease--is available. This review summarizes emerin and its emerging roles in nuclear "lamina" structure, chromatin tethering, gene regulation, mitosis, nuc...

Spatial and temporal control of signaling through lipid rafts.

Sphingolipid- and cholesterol-dependent microdomains (rafts) order proteins at biological membranes and have been implicated in most signaling processes at the cell surface, but the principles and mechanisms through which lipid rafts influence signaling are not well understood. Recent studies have revealed how lipid rafts are rapidly redistributed and assembled locally in response to extracellu...

Emery-Dreifuss muscular dystrophy mutations impair TRC40-mediated targeting of emerin to the inner nuclear membrane.

Emerin is a tail-anchored protein that is found predominantly at the inner nuclear membrane (INM), where it associates with components of the nuclear lamina. Mutations in the emerin gene cause Emery-Dreifuss muscular dystrophy (EDMD), an X-linked recessive disease. Here, we report that the TRC40/GET pathway for post-translational insertion of tail-anchored proteins into membranes is involved in...
